SESQUITERPENOIDS AS AVIAN MYELOBLASTOSIS VIRUS PROTEASE INHIBITORS PRODUCED BYStachybotrys sp

چکیده
Aspartic retroviral proteases play an important role in the processing of viral polyproteins into mature functional proteins1}. An inhibitor of avian myeloblastosis virus protease (AMV-protease) could be a possible candidate for antiviral-therapy and could be used, for example, as an anti-HIV agent2). In the course of screening for AMV-protease inhibitors, we discovered Mer-NF5003B, E and F, novel sesquiterpenoids (Fig. 1), from the culture broth of a fungal strain Stachybotrys sp. MerNF5003 (FERM p-12344). In this paper, the fermentation of the producing strain, isolation, physico-chemical properties, structure elucidation and the some biological activities of Mer-NF5003B, E and F are described. Screening of AMVprotease were carried out in the following manner: AMV-protease (Molecular Genetic Resources, 1 jug) was incubated with ThrPhe-Gln-Ala-Tyr-Pro-Leu-Arg-Glu-Ala (Peptide

Hyphodontal, a new antifungal inhibitor of reverse transcriptases from Hyphodontia sp. (Corticiaceae, Basidiomycetes).
In a search for inhibitors of RNA-directed DNA polymerases a new isolactarane sesquiterpenoid, hyphodontal (1), was isolated from fermentations of a Canadian Hyphodontia species. Its structure was elucidated by spectroscopic methods. Hyphodontal strongly inhibits the growth of several yeasts and is a non-competitive inhibitor of avian myeloblastosis virus (Ki 346 microM) and Moloney murine leuk...

DNA isolated from avian virus-producing leukemic myeloblasts induced the production of viruses, but not morphological transformation, in cultivated chicken fibroblasts. The recovered virus had the same biological characteristics as the original avian myeloblastosis virus (AMV) and produced myeloblastosis and nephroblastomas when injected into chickens. Neutralization experiments with chicken an...
متن کاملIsolation of infectious DNA from avian myeloblastosis virus transformed leukemic cells.
Chide helper factor (chf) negative chick embryo cells were exposed in the presence of DEAEdextran to chromosomal DNA isolated from avian leukemic myeloblasts which had been induced by infection with the BAI-A strain of avian myeloblastosis virus (AMV) (subgroup B ). After several transfers, avian C-type virus production was observed in the cultures with electron micro scopy, although no altera...
متن کاملDesign of new potent HTLV-1 protease inhibitors: in silico study
HTLV-1 and HIV-1 are two major causes for severe T-cell leukemia disease and acquired immune deficiency syndrome (AIDS). HTLV-1 protease, a member of aspartic acid protease family, plays important roles in maturation during virus replication cycle. The impairment of these proteases results in uninfectious HTLV-1virions.Similar to HIV-1protease deliberate mutations that confer drug resistance on...
متن کاملHigh molecular weight RNA from avian myeloblastosis virus.
Avian myeloblastosis virus produced in vitro by myeloblastic cells in suspension was labeled with 32P and 14Camino acids. After partial purification of the virus, and pronase digestion, an RNA was extracted by phenol into the aqueous phase. Glycerol gradient centrifugation showed that the 32P-labeled RNA con tained 2 distinct components: a high molecular weight fraction sedimenting at about 67 ...
